They should already be in iTunes or you'll at least get a message asking if you want to add to your iTunes library.

How to backup your iPod files to your local computer?
If you've bought it then make sure you're using the same iTunes Store account and you'll be able to redownload it for free if you need.

How to transfer your files from your old iPod to your new one?
Authorize the computer and then go to File-Transfer purchases from iTunes. Then take the music from the library and transfer to new iPod. Also, for non-iTunes music you can use Freecorder to record them off the old iPod into mp3's to put back into iTunes library.

How to transfer local files to iPod without iTunes?
I don't know how to do this in a way to make a song play but you can go to MyComputer or Computer from the Start menu and locate the iPod as it is basically a USB storage device.
